Tom Holland was not allowed to read the script for this movie, since he revealed too many secrets for Spider-Man: Homecoming (2017). In one of the promotional videos for the movie, several of its cast members are in an interrogation room refusing to reveal any spoilers, and Holland's mouth is conspicuously taped shut.
Mark Ruffalo revealed that he was given a fake script due to his habit of accidentally spoiling past Marvel movies. He jokingly claimed in an interview that the fake script was better than the real one.

Steve Rogers (Chris Evans) had a new look for this movie. At the end of Captain America: Civil War (2016), Steve Rogers quit as Captain America, something which occurred several times in the comic books. For the first time, he assumed the identity of "Nomad". Rogers' costume in this film captured basic elements of the Nomad character, in that it is primarily blue and features yellow, disk-like objects attached at the collar. For the second time Steve Rogers quit, he was featured to have grown scraggly with a beard, and he would eventually shave the beard and create a new identity as "The Captain". Consequently, the physical appearance and costume for Steve Rogers is a combination of the two most popular times he quit being Captain America.

Tony Stark (Robert Downey, Jr.) telling Peter Parker (Tom Holland) to cut it out with the pop culture references is quite hypocritical, as Stark often pokes fun at other characters by calling them by movie titles or character names. In The Avengers (2012), Stark called Loki Reindeer Games (2000), Rock of Ages (2012), and Shakespeare in the Park (1995), he called Thor Point Break (1991), and he called Hawkeye "Legolas". In Captain America: Civil War (2016), he called Bucky The Manchurian Candidate (2004). In Spider-Man: Homecoming (2017), he called Peter Parker "Crockett", as a reference to Miami Vice (1984). In this movie, he scorned Ebony Maw by calling him "Squidward", and he called Star Lord Flash Gordon (1980).

The poster contained a note from Mark Ruffalo explaining that the poster had not yet been released, and Holland was supposed to keep it a secret. Ironically, Holland had already showed the poster to the camera and read the message out loud before discovering a large sign on the back that read "CONFIDENTIAL, DO NOT SHARE". However, this seems like a deliberate set-up from the studio, as Holland and Ruffalo are known for revealing too many secrets about past Marvel movies. Plus, it lined up too conveniently with the release date of the first trailer. However, no proof exists.

This movie was inspired by the following Marvel Comics storylines: The Infinity Gauntlet (Thanos forms the Infinity Gauntlet and becomes all-powerful), The Thanos Imperative (the Guardians of the Galaxy take on Thanos), and Infinity (the Avengers and Inhumans team up to battle Thanos). The Marvel Comics storyline of the same name, however, involved a battle between superheroes for the Infinity Gauntlet, and has nothing to do with this story. This battle happened after the events of The Infinity Gauntlet and Infinity Crusade, where in the Infinity Gauntlet, Adam Warlock and his Infinity Watch formed by Gamora, Moondragon, Drax the Destructor, and Pip the Troll, allied forces with Thanos in order to defeat Magus, Warlock's evil side, who was looking to reunite the Infinity Gauntlet to be all-powerful and in the Infinity Crusade, Adam Warlock, The Avengers, Thanos, X-Men, the Fantastic Four, and Silver Surfer unite to defeat the Goddess, Warlock's good side, who was looking to convert the entire universe to worship her and control everyone.

The other role required him to have longer hair than he'd previously had as Spider-Man, so he had his stylists incorporate the new length into his look for this movie. However, after seeing the final product when the first trailer came out, Holland regretted this decision, saying he looked ridiculous and jokingly compared himself to Prince Charming.

In Thor (2011), Odin mentioned that Thor's hammer Mjölnir was "forged inside a dying star". Some fans took this as an indication that Mjölnir was made from neutronium, an extremely heavy substance that is sometimes left when big stars die (which would account for Mjölnir's extreme weight). The revelation that Mjölnir was forged in Nidavellir takes the metaphor even further, since Nidavellir is literally a forge inside the remains of a dead star.

Thanos' henchmen, known as "The Black Order", all have names that have something to do with blackness or darkness. Cull Obsidian, which basically means "black death". Ebony Maw, roughly "dark mouth". Proxima Midnight, or "among the darkness"/"of the darkness". Lastly, Corvus Glaive, whose first name is the name of the constellation depicting a raven. It's also the name for the genus of birds such as crows and ravens: black birds traditionally associated with death. So his name can be seen as black bird of death. Thor states he is one thousand five hundred-years-old, which gives his birthdate in the year 518 A.D. However, historically the first mention of Thor is (using the method of "interpretatio romana", where characteristics perceived to be similar by Romans result in identification of a Norse god as a Roman or Greek deity, in this case, Thor is equated to Hercules) by the Roman historian Tacitus is the first century (1 A.D. to 100 A.D.), predating the date Thor gave by four hundred years. Presumably though, after this amount of time, it would either be hard to remember exactly how many years you've lived, or it would be unimportant. Thor may have said fifteen hundred years for an "at least" approximation.

During the post-credits scene of Avengers: Age of Ultron (2015), Thanos grabs a Gem-less Infinity Gauntlet and says, "Fine, I'll do it myself." An Infinity Gauntlet has been seen in the Marvel Cinematic Universe before, as it can be seen being held in the weapons vault on Asgard in Thor (2011). In a recent interview with Marvel Studios President Kevin Feige, he was asked if the post-credits scene in Avengers: Age of Ultron (2015) was made to fix the continuity error of the Gauntlet being spotted in Thor (2011). He stated "No", as there is in fact more than one Infinity Gauntlet in the Marvel Cinematic Universe. The Gauntlet in Asgard was called a fake by Hela in Thor: Ragnarok (2017). In addition, this movie's revelation that Thanos' Infinity Gauntlet was formed at Nidavellir, the same place where Asgard got most of its weapons, could potentially explain a relationship between the two gauntlets.

There is no mid credits scene (nothing comes after the principal actors and actresses names are shown at the end of the movie), unlike most previous Marvel Cinematic Universe movies. This was done intentionally by the filmmakers to present a feeling of hopeless for the heroes and the audience in light of the tragedy for the entirety of the credits. As a contrast, the post credit scene is meant to serve as a glimmer of hope without presenting that everything will be fixed later.

When he chokes Loki to death, Thanos says "No resurrections this time." This is likely an inside joke referring to how Loki had already faked his death twice: He fell away into space in Thor (2011) only to show up again in The Avengers (2012), and he created the illusion of being stabbed in Thor: The Dark World (2013) so that he could return to Asgard and dethrone Odin. However, this death is real, and is most likely permanent unless something happens in Avengers: Endgame (2019).

When Gamora and Thanos travel to Vormir to obtain the Soul Stone, they meet the guardian of the gem, who is revealed to be Johann Schmidt (Red Skull). It confirms the seven-years-ongoing fan theory that the Tesseract didn't kill Schmidt at the end of Captain America: The First Avenger (2011), but had just sent him off to space. It seems fitting that, as he was consumed by the power of the Space Stone (Tesseract) in that movie, he would be the guardian of the Soul Stone in this movie, being his first appearance since then. However, Hugo Weaving did not reprise his role from Captain America: The First Avenger (2011) due to previous reluctance to return to portray the character and the long sessions to apply make-up as well, leading him to be replaced by Ross Marquand .

The Russo Brothers stated in a podcast interview, that the actual reason why The Incredible Hulk refuses to, "come out", wasn't because he's scared of Thanos, (while he certainly was taken back a bit by being so easily defeated by him in the beginning), but rather The Incredible Hulk simply being tired of Bruce Banner only using him, when he needs him during a battle and refusing so to spite him.

Thanos' mass genocide at the end affects mainly later Avenger members, like Bucky Barnes, Falcon, Spider-Man, Doctor Strange, Black Panther, and the Scarlet Witch. The original Avengers members (Iron Man, Captain America, The Incredible Hulk, and Black Widow) are still alive, with only Hawkeye unaccounted for as he does not appear in this movie. In contrast, the original members of the Guardians of the Galaxy are all wiped out with the exception of Rocket (and Gamora, who had already died earlier).

The post credits scene contains another Easter egg reference to Samuel L. Jackson outside of the Marvel Cinematic Universe. As he's being wiped from existence, Nick Fury mutters "Motherf-----". Jackson is known professionally for portraying characters who use that expletive profusely, and has a huge cult following as a result. In Captain America: The Winter Soldier (2014), "The path of the righteous man: Ezekiel 25:17" can be seen on Fury's grave stone, a reference to a monologue he delivered in Pulp Fiction (1994) as Jules Winnfield, one of his most famous and profane roles.

Multiple trailers for the movie featured a shot of The Incredible Hulk running along with other Avengers and Wakandans, but this does not occur in the actual movie. Several instances of Bruce Banner attempting to transform into The Incredible Hulk after his fight with Thanos fail specifically because The Incredible Hulk personality is resisting.

In the teaser trailer for this movie, shown at a San Diego Comic-Con, the shot seen before transitioning to the title card shows Thanos with a fully assembled Infinity Gauntlet. In Avengers: Age of Ultron (2015), we get another look at the Gauntlet in the form of an ethereal image with four Infinity Stones set in their respective positions. With the red, blue, yellow, and purple Infinity Stones revealed and identified as Reality, Space, Mind, and Power, it is safe to assume that the orange gem set in the center of the Gauntlet and the green gem positioned atop one of the knuckles, are the Infinity Stones of Soul and Time respectively. In this movie, however, when Thanos completes the Gauntlet, the Mind Stone is in the center, the Stones of Power, Space, Reality, and Soul are on the knuckles, and the Time Stone is on the thumb.

There's a deleted scene in the Blu-ray copy of the movie that explains why The Guardians are on Titan. In the movie, the last time we see them before the scene on Titan is when they go to Knowhere to get the Reality Stone, only to realize that Thanos already has it, and is waiting for Gamora, as she has information on the Soul Stone, and he leaves with her. In the Blu-ray copy, Nebula dispatches the guard and frees herself. Then she goes to computer terminal and sends a message to Mantis telling the team to meet her in Titan. This scene was not a separate deleted scene, but incorporated in the movie.

The villains repeatedly threaten the heroes with murder of their associates: 1) Thanos threatens to kill Thor if Loki does not hand over the tesseract (Space Stone). 2) The Black Order offer to spare Scarlet Witch if The Vision surrenders and gives up the Mind Stone. 3) Thanos tortures and threatens to kill Nebula if Gamora does not reveal the location of the Soul Stone. 4) As recounted by Eitri, he was forced to forge the Infinity Gauntlet by Thanos under the threat of having his fellow dwarves murdered. 5) Although not explicitly asked to do so by Thanos, Doctor Strange gives up the Time Stone in exchange for Tony Stark's life.

When Nat and T'Challa face Proxima Midnight and Cull Obsidian, with Wakanda's force field between them, T'Challa says when the battle is over, what's left will be "dust, and blood". He was not wrong about the dust. Doctor Strange tells Tony before their battle that if it comes down to saving their lives or the Time Stone, he would always pick the Time Stone, he later uses the Stone to look into the future, noting that in the over fourteen million possible futures he saw, there was only one scenario where they win, later during the battle, he sacrifices the Time Stone to save Tony, and right before he's disintegrated by Thanos, he says, "There was no other way". If he's telling the truth in all of these instances, it all points to a long game of that one possible future, being one where he sacrifices the Time Stone first, so that it will ultimately be saved later. One of Drax's first scenes in the movie is a gag where he thinks he can make himself disappear, Ned Leeds' only line in the movie is the entirely accurate, "WE'RE ALL GONNA DIE!", T'Challa warns Proxima Midnight that "Thanos will have nothing but, "dust and blood", before the battle, Tony Stark provides the blood on Titan, Thanos provides the dust, killing half of the population in the galaxy.

Rocket's survival was foreshadowed earlier in the movie, when Thor asks if Rocket (rabbit) is the Captain, to which Rocket sarcastically says he is to spite Quill, "foreshadowing" the ending when all of the Guardians disintegrate except him, ironically making him the Captain, and when he and Thor are discussing loss, Thor says something to the effect of having nothing left to lose, and Rocket mumbles (twice),- "I've got some things to lose." Losing all of his friends, his family, including Groot.

At one point, Doctor Strange uses the Time Stone to look ahead and see all possible futures regarding their war with Thanos. This is undoubtedly a tactic he had learned from the Ancient One. In Doctor Strange (2016), she had foreseen all of her own possible futures (which all lead to her death and no further) as well as those of Doctor Strange.
